(1) Background: In hospitals, medical and dental clinics, antiseptics or disinfectants play an essential role in the control of nosocomial infections. This study aimed to evaluate R. officinalis and P. paniculata glycolic extracts regarding: (I) their antimicrobial action on planktonic and biofilm (monotypic and cutaneous biofilm model-S. aureus, S. epidermidis and C. acnes); and (II) their cytotoxicity on human keratinocytes (HaCaT). (2) Methods: Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were performed (CLSI protocol M7-A6 and M11-A8). MTT analysis was used to evaluate the antibiofilm activity of the extracts on biofilms and their cytotoxicity on human keratinocytes. (3) Results: The combined glycolic extracts MIX A (75% P. paniculata + 25% R. officinalis); MIX B (50% P. paniculata + 50% R. officinalis); and MIX C (25% P. paniculata + 75% R. officinalis) promoted MBC values by 50 mg/mL on S. aureus, absent on S. epidermidis, and ranged 6.25-50 mg/mL for C. acnes. The cutaneous biofilm model was reduced more than 90%. In addition, it showed biocompatibility with human keratinocytes, resulting in percentages of viability greater than 50%. (4) Conclusions: The combination of extracts promoted antimicrobial action on planktonic cultures, and monotypic and heterotypic biofilms of skin pathogens. Additionally, these extracts are biocompatible against human keratinocytes.

Objetivo:Relatar a experiência de uma equipe de enfermeiros estomaterapeutas na construção de um algoritmo para a indicação de equipamento coletor para estomias de eliminação. Método: Relato de experiência, do período de janeiro de 2018 a setembro de 2019, sobre o processo de construção de um algoritmo para indicação de equipamento coletor para estomias de eliminação. Resultados: A partir de determinadas características clínicas (parâmetros de avaliação) e da categorização dos equipamentos coletores (solução), foi desenvolvido um algoritmo para indicação de equipamento coletor para estomias de eliminação. Conclusão: Espera-se que esse instrumento possa auxiliar os enfermeiros na sua prática profissional quanto à escolha do equipamento coletor e na construção de protocolos clínicos.

Conclusion: It is expected that this instrument can help nurses in their professional practice regarding the choice of collecting equipment and the construction of clinical protocols.

Conclusión: Se espera que este instrumento pueda ayudar a los enfermeros en su práctica profesional en cuanto a la elección de equipos de recolección y la construcción de protocolos clínicos.

Biomodified coated-lipid vesicles were obtained using the DPPC lipid (L) and F127 copolymer linked covalently with spermine (SN), biotin (BT), and folic acid (FA), resulting in LF127-SN, LF127-BT, and LF127-FA nanoplatforms. The photosensitizer hypericin (HY) was incorporated into the nanosystem by a thin-film method and characterized by dynamic light scattering, zeta potential, encapsulation efficiency, and transmission electronic microscopy. The results provided a good level of stability for all nanoplatforms for at least 5 days as an aqueous dispersion. The in vitro serum stability showed that the HY-loaded LF127-SN has a lower tendency to form complexes with BSA protein than with its analogs. LF127-SN was the most stable HY formulation, followed by LF127-BT and LF127-FA, confirmed by the association constant (Kd) values: 600 µmol L-1, 1100 µmol L-1, 515 µmol L-1, and 378 µmol L-1 for LF127, LF127 FA, LF127-BT, and LF127-SN, respectively. The photodynamic potential of HY was accessed by cytotoxicity assays using Caco-2, B16-F10, L-929, and HaCat cells. HY-loaded LF127-SN revealed a significant increase in the selectivity compared to other nanoplatforms. HY-loaded in LF127-BT and LF127-SN showed distinct uptake and biodistribution after 2 h of intravenous application. All biomodified coated-lipids showed satisfactory metabolism within 72 h after application, without significant accumulation or residue in any vital organ. These results suggest that incorporating HY-loaded in these nanosystems may be a promising strategy for future applications, even with a small amount of binders to the coating copolymer (0.02% w/v). Furthermore, these results indicate that the LF127-SN showed remarkable superiority compared to other evaluated systems, being the most distinct for future photodynamic therapy and theranostic applications.

Copaiba oil-resin (COR) extracted from Copaifera reticulata Ducke has been used as a natural chemotherapeutic agent for a wide range of therapeutic applications. This study presents an emulgel design with a high concentration of COR, designed to prevent and treat mastitis. The COR was stabilized in a gel matrix constituted by carbopol C934P and Pluronic® F127 (ECO formulation) ratios. The permeation study of ECO was accessed by Fourier transform infrared photoacoustic spectroscopy (FTIR-PAS). The results reveal a high capacity of ECO to permeate deep skin layers. Dairy cows with a history of mastitis were used as in vivo models and exposed to ECO treatment. Monitoring of the teat's inflammatory response showed that ECO effectively prevents mastitis. Furthermore, the ECO formulation was able to form a thin film gel on the application side, preventing fly proliferation and significantly reducing the pathogen load. This study reveals a drug that can used as an alternative application for mastitis in human or veterinary clinics.

Good management practices such as post-dipping applications (post-milking immersion bath) contribute to the dairy cattle health during lactation and minimize the appearance of mastitis (an infection in the mammary gland). The post-dipping procedure is performed conventionally using iodine-based solutions. The search for therapeutic modalities that are not invasive and do not cause resistance to the microorganisms that cause bovine mastitis instigates the interest of the scientific community. In this regard, antimicrobial Photodynamic Therapy (aPDT) is highlighted. The aPDT is based on combining a photosensitizer (PS) compound, light of adequate wavelength, and molecular oxygen (3O2), which triggers a series of photophysical processes and photochemical reactions that generate reactive oxygen species (ROS) responsible for the inactivation of microorganisms. The present investigation explored the photodynamic efficiency of two natural PS: Chlorophyll-rich spinach extract (CHL) and Curcumin (CUR), both incorporated into the Pluronic® F127 micellar copolymer. They were applied in post-dipping procedures in two different experiments. The photoactivity of formulations mediated through aPDT was conducted against Staphylococcus aureus, and obtained a minimum inhibitory concentration (MIC) of 6.8 mg mL-1 for CHL-F127 and 0.25 mg mL-1 for CUR-F127. Only CUR-F127 inhibited Escherichia coli growth with MIC 0.50 mg mL-1. Concerning the count of microorganisms during the days of the application, a significant difference was observed between the treatments and control (Iodine) when the teat surface of cows was evaluated. For CHL-F127 there was a difference for Coliform and Staphylococcus (p < 0.05). For CUR-F127 there was a difference for aerobic mesophilic and Staphylococcus (p < 0.05). Such application decreased bacterial load and maintained the milk quality, being evaluated via total microorganism count, physical-chemical composition, and somatic cell count (SCC).

In this study, we developed a bioadhesive emulsion-filled gel containing a high amount of Copaifera reticulata Ducke oil-resin as a veterinary or human clinical proposal. The phytotherapeutic system had easy preparation, low cost, satisfactory healing ability, and fly repellency, making it a cost-effective clinical strategy for wound care and myiasis prevention. Mechanical, rheological, morphological, and physical stability assessments were performed. The results highlight the crosslinked nature of the gelling agent, with three-dimensional channel networks stabilizing the Copaifera reticulata Ducke oil-resin (CrD-Ore). The emulgel presented antimicrobial activity, satisfactory adhesion, hardness, cohesiveness, and viscosity profiles, ensuring the easy spreading of the formulation. Considering dermatological application, the oscillatory responses showed a viscoelastic performance that ensures emulgel retention at the action site, reducing the dosage frequencies. In Vivo evaluations were performed using a case report to treat ulcerative skin wounds aggravated by myiasis in calves and heifers, which demonstrated healing, anti-inflammatory, and repellent performance for the emulsion-filled gel. The emulgel preparation, which is low in cost, shows promise as a drug for wound therapy.

INTRODUCTION: Given the advances in diagnosis and clinicalsurgical therapies, congenital heart defects are increasing their prevalence in the adult population. Yet, there is still no consensus on the best way to follow up with these patients. Among the validated tools for monitoring patients with heart disease, the Cardiopulmonary Exercise Test (CPET) plays an important role. However, the use of this test for monitoring the population of Grown Up Congenital Heart (GUCH) still lacks studies. This study aimed to assess the correlation between ergospirometry variables and the severity of the GUCH population measured by echocardiographic aspects. METHODS: A retrospective cohort with 248 GUCH (Table 1) over 18 years old, was referred to CPET, from 2015 to 2021, in a tertiary hospital in the state of São Paulo. Patients were sequentially included, and those in functional class IV or with contraindications to CPET were excluded. Ergospirometry variables were analyzed in association with ventricular function - estimated by echocardiogram in the same period. RESULTS: Most patients were in functional class I and II (86.3%). In the echocardiographic findings, 40% had pulmonary hypertension and almost all had preserved left ventricle function. The CPET showed a median VO2 peak around 69% of predict. Other parameters are summarized in Table 2. CPET variables were able to stratify the severity of GUCH, mainly by pulmonary hypertension. Comparing CPET data and imaging diagnosis, VE/VCO2 slope> 32 and OUES <60% were related to the presence of pulmonary hypertension. CONCLUSION: The CPET is an important resource for prognostic and diagnostic definition in the evolution of GUCH patients.

This work aims at the design and engineering of sustainable biomaterials based on natural fibers to replace non-renewable fiber sources in the development of non-woven delivery systems. Cellulose fibers were used as the main support to produce multi-structured materials with the incorporation of microfibrillated cellulose (MFC) as an additive. A 3D carboxymethylcellulose matrix retaining a natural bioactive product, eucalyptus essential oil, (CMC/EO), with controlled release functionalities, was also applied to these materials using bulk and spray coating methodologies. Additionally, using a 3D modeling and simulation strategy, different interest scenarios were predicted to design new formulations with improved functional properties. Overall, the results showed that MFC provided up to 5% improved strength (+48%) at the expense of reduced softness (-10%) and absorbency (-13%) and presented a good potential to be used as an additive to maximize natural eucalyptus fibers content in formulations. The addition of CMC/EO into formulations' bulk revealed better strength properties (21-28%), while its surface coating improved absorption (23-25%). This indicated that both application methods can be used in structures proposed for different sustainable applications or a more localized therapy, respectively. This optimization methodology consists of a competitive benefit to produce high-quality functionalized biomaterials for added-value applications.

Multifunctional P123 micelle linked covalently with spermine (SM) and folic acid (FA) was developed as a drug delivery system of hypericin (HYP). The chemical structures of the modified copolymers were confirmed by spectroscopy and spectrophotometric techniques (UV-vis, FTIR, and 1H NMR). The copolymeric micelles loading HYP were prepared by solid dispersion and characterized by UV-vis, fluorescence, dynamic light scattering (DLS), ζ potential, and transmission electron microscopy (TEM). The results provided a good level of stability for HYP-loaded P123-SM, P123-FA, and P123-SM/P123-FA in the aqueous medium. The morphology analysis showed that all copolymeric micelles are spherical. Well-defined regions of different contrast allow us to infer that SM and FA were localized on the surface of micelles, and the HYP molecules are located in the core region of micelles. The uptake potential of multifunctional P123 micelle was accessed by exposing the micellar systems loading HYP to two cell lines, B16-F10 and HaCaT. HYP-loaded P123 micelles reveal a low selectivity for melanoma cells, showing significant photodamage for HaCat cells. However, the exposition of B16-F10 cells to Hyp-loaded SM- and FA-functionalized P123 micelles under light irradiation revealed the lowest CC50 values. The interpretation of these results suggested that the combination of SM and FA on P123 micelles is the main factor in enhancing the HYP uptake by melanoma cells, consequently leading to its photoinactivation.

The growing demand for tissue papers worldwide encourages the paper industry to find new approaches to optimize the raw materials furnish management, and simultaneously to improve tissue paper performance. Softness, strength, and absorption are the key tissue properties that enhance the attention of both industry and consumers. Fiber morphology, fiber modification process steps, and structural properties affect these functional properties, and, therefore, the efforts to evaluate them and establish the relationship or models that describe them constitute a multifactorial challenge. For this purpose, we aimed to investigate the trade-off between the input variables (morphological, suspension, and structural properties) and the final properties. Key variables like the type of furnish raw materials, including the fiber mixture, mechanical and enzymatic treatments, additives incorporation, and the type of industrial base tissue papers were taken under consideration. To achieve these relationships, we used different data-driven modeling approaches including multiple linear regression (MLR), artificial neural networks (ANN), and a 3D fiber-based simulator. The MLR and ANN models were built by data collected from an experimental design, and isotropic laboratory structures were prepared and tested for changes in structural and functional properties. Moreover, a 3D fiber-based simulator was used to investigate the influence of fibers on structural properties. These results indicated that the realistic predictions enabled us to link fiber and tissue structure characteristics. In conclusion, this work has revealed that this computational modeling approach can be used to model the effect of fiber pulps parameters with final end-use tissue properties, allowing to design innovative tissue products.

Pluronic/lipid mix promises stealth liposomes with long circulation time and long-term stability for pharmaceutical applications. However, the influence of Pluronics on several aspects of lipid membranes has not been fully elucidated. Herein it was described the effect of Pluronics on the structured water, alkyl chain conformation, and kinetic stability of dimyristoylphosphatidylcholine (DMPC) liposomes using interfacial and deeper fluorescent probes along with computational molecular modeling data. Interfacial water changed as a function of Pluronics' hydrophobicity with polypropylene oxide (PPO) anchoring the copolymers in the lipid bilayer. Pluronics with more than 30-40 PO units had facilitated penetration at the bilayer while shorter PPO favored a more interfacial interaction. Low Pluronic concentrations provided long-term stability of vesicles by steric effects of polyethylene oxide (PEO), but high amounts destabilized the vesicles as a sum of water-bridge cleavage at the polar head group and the reduced alkyl-alkyl interactions among the lipids. The high kinetic stability of Pluronic/DMPC vesicles is a proof-of-concept of its advantages and applicability in nanotechnology over conventional liposome-based pharmaceutical products for future biomedical applications.

Biotin, spermine, and folic acid were covalently linked to the F127 copolymer to obtain a new drug delivery system designed for HY-loaded PDT treatment against B16F10 cells. Chemical structures and binders quantification were performed by spectroscopy and spectrophotometric techniques (1NMR, HABA/Avidin reagent, fluorescamine assay). Critical micelle concentration, critical micelle temperature, size, polydispersity, and zeta potential indicate the hydrophobicity of the binders can influence the physicochemical parameters. Spermine-modified micelles showed fewer changes in their physical and chemical parameters than the F127 micelles without modification. Furthermore, zeta potential measurements suggest an increase in the physical stability of these carrier systems. The phototherapeutic potential was demonstrated using hypericin-loaded formulation against B16F10 cells, which shows that the combination of the binders on F127 copolymer micelles enhances the photosensitizer uptake and potentializes the photodynamic activity.

Tissue paper production frequently combines two main types of raw materials: cellulose fibers from renewable sources and polymer-based additives. The development of premium products with improved properties and functionalities depends on the optimization of both. This work focused on the combination of innovative experimental and computational strategies to optimize furnish. The main goal was to improve the functional properties of the most suitable raw materials for tissue materials and develop new differentiating products with innovative features. The experimental plan included as inputs different fiber mixtures, micro/nano fibrillated cellulose, and biopolymer additives, and enzymatic and mechanical process operations. We present an innovative tissue paper simulator, the SimTissue, that we have developed, to establish the correlations between the tissue paper process inputs and the end-use paper properties. Case studies with industrial interest are presented in which the tissue simulator was used to design tissue paper materials with different fiber mixtures, fiber modification treatments, micro/nano fibrillated cellulose, and biopolymer formulations, and to estimate tissue softness, strength, and absorption properties. The SimTissue was able to predict and optimize a broader range of formulations containing micro/nanocellulose fibers, biopolymer additives, and treated-fiber mixtures, saving laboratory and industrial resources.

The furnish management of tissue materials is fundamental to obtain maximum quality products with a minimum cost. The key fiber properties and fiber modification process steps have a significant influence on the structural and functional properties of tissue paper. In this work, two types of additives, a commercial biopolymer additive (CBA) that replaces the traditional cationic starch and micro/nanofibrillated cellulose (CMF), were investigated. Different formulations were prepared containing eucalyptus fibers and softwood fibers treated mechanically and enzymatically and both pulps with these two additives incorporated independently and simultaneously with drainage in the tissue process range. The use of these additives to reduce the percentage of softwood fibers on tissue furnish formulations was investigated. The results indicated that a maximum of tensile strength was obtained with a combination of both additives at the expense of softness and water absorbency. With a reduction of softwood fibers, the incorporation of additives increased the tensile strength and water absorbency with a slight decrease in HF softness compared with a typical industrial furnish. Additionally, a tissue computational simulator was also used to predict the influence of these additives on the final end-use properties. Both additives proved to be a suitable alternative to reduce softwood fibers in the production of tissue products, enhancing softness, strength and absorption properties.

The mode-mismatched dual-beam thermal lens technique is widely applied in the characterization of optical and thermo-physical properties of solids and liquids. The technique has also been used to investigate transient acoustic waves induced by pulsed laser excitation at the nanosecond time scale. In this paper, we developed a semi-analytical model to describe the transient acoustic wave that allows a fitting procedure to get the physical properties of fluid samples. The method was used to investigate samples with different mixtures of ethanol and water, and quantitative information of piezo-optic coefficient and sound speed are evaluated for the fluid mixtures.

Phthalocyanine aluminum chloride (Pc) is a clinically viable photosensitizer (PS) to treat skin lesions worsened by microbial infections. However, this molecule presents a high self-aggregation tendency in the biological fluid, which is an in vivo direct administration obstacle. This study proposed the use of bioadhesive and thermoresponsive hydrogels comprising triblock-type Pluronic F127 and Carbopol 934P (FCarb) as drug delivery platforms of Pc (FCarbPc)-targeting topical administration. Carbopol 934P was used to increase the F127 hydrogel adhesion on the skin. Rheological analyses showed that the Pc presented a low effect on the hydrogel matrix, changing the gelation temperature from 27.2 ± 0.1 to 28.5 ± 0.9 °C once the Pc concentration increases from zero to 1 mmol L-1. The dermatological platform showed matrix erosion effects with the release of loaded Pc micelles. The permeation studies showed the excellent potential of the FCarb platform, which allowed the partition of the PS into deeper layers of the skin. The applicability of this dermatological platform in photodynamic therapy was evaluated by the generation of reactive species which was demonstrated by chemical photodynamic efficiency assays. The low effect on cell viability and proliferation in the dark was demonstrated by in vitro assays using L929 fibroblasts. The FCarbPc fostered the inhibition of Staphylococcus aureus strain, therefore demonstrating the platform's potential in the treatment of dermatological infections of microbial nature.

Hypericin (Hy) compound presents a high photoactivity in photodynamic therapy (PDT), photodiagnosis and theranostics applications. The maintenance of this compound in monomeric form could undermine the potential benefits of its photophysical and photodynamic activity. In this study, we demonstrated that the Hy formulated in a system based on the use of the F127 copolymer and the 1,2-dipalmitoyl-sn-3-glycerol-phosphatidylcholine (DPPC) as micelles, liposomal vesicles and Copolymer-Lipid coated systems, have improved its photophysical properties for many clinical modalities. Based on the results of the triplet state lifetime values (τt), the singlet oxygen quantum yield (ΦΔ1O2), the fluorescence lifetime (τF) and the fluorescence quantum yield (ΦF), all Hy formulations had its photophysical properties described in different models of drug delivery systems (DDS). In addition, the transient spectra profile of those formulations was unaffected by the Hy incorporation process, except for the liposomal system, which demonstrated to be the less stable one by flash photolysis technique. The cytotoxic effects of those formulations were also investigated for CaCo-2 and HaCat cells line. The cytotoxic concentrations for 50% (CC50) were 0.56, 1.05, 1.33 and 4.80 µmol L-1 for Copolymer-Lipid/Hy, DPPC/Hy, F127/Hy and ethanol/Hy for CaCo-2 cells, respectively, and 0.69, 2.02, 1.45 and 1.16 µmol L-1 for Copolymer-Lipid/Hy, DPPC/Hy, F127/Hy and ethanol/Hy for HaCat cells, respectively. The F127 copolymer had a significant role in many photophysical parameters determined for Copolymer-Lipid/Hy coated system. Although all those formulations had shown satisfactory results, Copolymer-Lipid/Hy proved to be superior in many aspects, being the most promising formulation for PDT, photodiagnosis and theranostics applications.

Cosmetic products in which all the skincare compounds are biomolecules, biocompatible and biodegradable constitute a request of an educated consumer corresponding to a premium cosmetic segment. For this purpose, a cellulose-based delivery system was developed to retain biomolecules for dermic applications. The 3D matrix was built with microfibrillated cellulose, nanofibrillated cellulose and carboxymethylcellulose combined with a crosslinking agent, the alginate, to obtain a 3D matrix capable of retaining and releasing bioactive components of microalgae Chlorella vulgaris and tea tree essential oil. The porosity and pore dimensions and uniformity of this support matrix were optimized using 3D computational tools. The structures of the biopolymers were characterized using SEM, EDX, FTIR-ATR and DSC techniques. The essential oil and the microalgae components were successfully incorporated in a 3D stable matrix. The results indicate that the polymeric matrix retains and releases the essential oil biomolecules in a controlled way, when compared with tea tree essential oil, which is vaporized from 25 °C to 38 °C, without this 3D polymeric matrix. The microalgae and cellulose-based delivery system proved to be an interesting option for dermic and cosmetic applications because the exposure time of the therapeutic biomolecules was improved, and this factor consists of a competitive benefit for dermic systems.

Tissue paper consumption has been growing for the past years, with a forecasted increase in demand for premium products. Premium tissue paper products are obtained with a balance among softness, strength, and absorption properties, optimized for each kind of tissue paper. These properties are influenced by the three-dimensional structure, made from the spatial distribution of cellulose fibres. To our knowledge, the efforts made to date to improve the softness, strength and absorption properties have overlooked the 3D structure. There is an absence of 3D experimental data in the literature for the simultaneous characterization of individual eucalyptus fibres and the paper structure made from these fibres. The 2D fibre morphology determination, including fibre length and fibre width, was obtained by an image analysis method for pulp fibre suspensions, using the MorFiⓇ equipment. The third fibre dimension, the fibre thickness morphology in the out-of-plane direction, was obtained using SEM images of non-pressed isotropic laboratory-made paper sheets. The effective fibre thickness morphology, consisting of the fibre wall and lumen, was measured in the paper structure, as this is precisely the key fibre parameter, influencing not only the structure-related properties, such as paper thickness, bulk, and porosity, but also the final end-use properties. The paper structures were produced using an ISO standard adapted method, for tissue paper structures, without pressing, with a basis weight range from 20 to 150 g/m2. These data are important, among other possible uses, for paper property optimization and simulation studies with 3D fibre based simulators.

Hypericin (Hyp) is considered a promising photosensitizer for Photodynamic Therapy (PDT), due to its high hydrophobicity, affinity for cell membranes, low toxicity and high photooxidation activity. In this study, Hyp photophysical properties and photodynamic activity against melanoma B16-F10 cells were optimized using DPPC liposomes (1,2-dipalmitoyl-sn-glycero-3-phosphocholine) as a drug delivery system. This nanoparticle is used as a cell membrane biomimetic model and solubilizes hydrophobic drugs. Hyp oxygen singlet lifetime (τ) in DPPC was approximately two-fold larger than that in P-123 micelles (Pluronic™ surfactants), reflecting a more hydrophobic environment provided by the DPPC liposome. On the other hand, singlet oxygen quantum yield values (ΦΔ1O2) in DPPC and P-123 were similar; Hyp molecules were preserved as monomers. The Hyp/DPPC liposome aqueous dispersion was stable during fluorescence emission and the liposome diameter remained stable for at least five days at 30 °C. However, the liposomes collapsed after the lyophilization/rehydration process, which was resolved by adding the lyoprotectant Trehalose to the liposome dispersion before lyophilization. Cell viability of the Hyp/DPPC formulation was assessed against healthy HaCat cells and high-metastatic melanoma B16-F10 cells. Hyp incorporated into the DPPC carrier presented a higher selectivity index than the Hyp sample previously solubilized in ethanol under the illumination effect. Moreover, the IC50 was lower for Hyp in DPPC than for Hyp pre-solubilized in ethanol. These results indicate the potential of the formulation of Hyp/DPPC for future biomedical applications in PDT treatment.
